Akt, also known as protein kinase B (PKB), plays important role in cell survival, growth, proliferation, metabolism and migration. Overexpression of phosphorylated Akt was detected in tumor cells, which promoted excessive proliferation, anti-apoptosis, enhanced metastasis of these cells. Akt signaling pathway associated protein was considered important targets for interventional therapy of cancer. And numerous protein-targeted drugs have been used in clinical trials. To date, Akt signaling pathway is still the target for drug development. Although Akt signaling pathway showed gradual increase throughout tumorigenesis of melanoma (i.e. from normal melanocytes to melanoma as well as malignant melanoma), its mechanism was still not clear. In this study, we aim to investigate the regulatory mechanism of Akt signaling pathway during malignant transformation of melanoma cells, and screen a compound for the treatment of melanoma by inhibition of Akt signaling pathway.

Akt(蛋白激酶B)信号通路是调节细胞存活、生长、增殖、代谢和迁移等方面的主要通路,越来越多的研究表明,在多种肿瘤细胞中Akt信号经常失调,导致磷酸化Akt的过量表达,从而促进了肿瘤细胞的过度增殖、抗凋亡和转移性加强等性状。临床上,参与Akt信号的蛋白是多种肿瘤干预治疗的重要靶标。事实上,一些以这些蛋白为肿瘤治疗靶点的药物已经进入了临床试验阶段,并且,Akt信号仍然是肿瘤药物设计的重要目标。在黑色素肿瘤的形成过程中,即从正常黑素细胞到黑色素瘤细胞,再到恶性黑色素瘤细胞,Akt通路活性逐渐增加,但是其具体机制仍不明了。本文就黑色素瘤细胞恶性转变过程中Akt信号的调控机制进行了深入的探讨,并筛选出通过抑制Akt信号而发挥作用的高效的治疗黑色素瘤的化合物。
