  本研究分别对大豆细胞质雄性不育系NJCMS2A及其保持系NJCMS2B的基因组DNA和转录组cDNA的atp9 基因进行PCR扩增、克隆和测序分析,发现从基因组DNA中扩增的atp9 基因序列均保持一致,而以转录组cDNA为模板扩增的atp9 基因的序列存在差异。生物信息学预测结果表明不育系NJCMS2A中atp9基因与保持系NJCMS2B相比,在转录过程中没有经过RNA编辑,使翻译后蛋白跨膜方向发生改变,从而导致蛋白结构不稳定,使得植株不能正常生长发育,出现败育现象。至于atp9基因的RNA编辑与大豆细胞质雄性不育的关系,还有待于进行转基因功能验证。

  In this report, PCR amplification, cloning and sequencing analysis about the genomic DNA and the transcription cDNA of atp9 gene in CMS line NJCMS1A and its maintainer line NJCMS1B were performed in soybean respectively. The results showed the sequences of atp9 gene amplified from genomic DNA were consistent, but amplified from transcriptomic cDNA were different. No RNA editing was performed in the transcription process of atp 9 gene in CMS line NJCMS1A compared with that of its maintainer line NJCMS1B according to bioinformation prediction. Trans-membrane orientation changed after protein translation, leading to unstable protein structure, abnormal growth and sterility in plants. With regards to the relation between atp 9 gene of RNA editing and CMS in soybean, further transgene function verification was needed.
